Pathology · Inflammation and repair: P-selectin mobilisation
Sample itemWithin minutes of exposure to histamine, venular endothelium shows a marked increase in leukocyte rolling. Which change at the endothelial surface explains this rapid response?
Original practice item, not a recalled exam question. Nothing here is recorded.
The answer and the reasoning
C. Redistribution of P-selectin from Weibel-Palade bodies to the surface
Mediators such as histamine and thrombin cause P-selectin stored in Weibel-Palade bodies to move to the endothelial surface within minutes. E-selectin and ICAM-1 appear later, over one to two hours, when IL-1 and TNF induce their transcription. Selectins bind sialylated oligosaccharides on leukocytes to slow them.
Option by option
A. Shedding of L-selectin from the endothelium
L-selectin is expressed on leukocytes, not on endothelium.
B. Increased affinity of integrins for ICAM-1
Integrin affinity changes occur on leukocytes, mediated by chemokines, after rolling.
C. Redistribution of P-selectin from Weibel-Palade bodies to the surface (correct)
Correct. Stored P-selectin is mobilised within minutes without new synthesis.
D. New transcription of E-selectin through NF-kB
E-selectin needs gene transcription and appears after one to two hours.
